We present a single-nucleus RNA sequencing (snRNA-seq) dataset of human skeletal muscle derived from 16 donors in the Singapore Adult Metabolism Study (SAMS1&2) cohort. The samples were collected from vastus lateralis muscle biopsies of individuals across multiple Asian ancestries (Chinese, Malay, and Indian), encompassing both lean and obese metabolic states, as well as before and after weight-loss. This dataset captures the cellular heterogeneity of human skeletal muscle and provides a resource for investigating ancestry-specific and metabolic state-associated transcriptional variation. In particular, the inclusion of diverse Asian populations addresses a critical gap in current human muscle transcriptomic references, which are predominantly derived from European cohorts.

Single-nucleus RNA-seq of Asian Skeletal Muscle Reveals Ancestry-specific Regulatory Programs Across Metabolic States

This dataset consists of skeletal muscle snRNA-seq data from two cohorts: SAMS1 (cross-sectional) and SAMS2 (longitudinal weight-loss study). It includes samples from three Asian ancestries (Chinese, Malay, and Indian), comprising 8 lean, 4 obese individuals, and 6 samples from the longitudinal cohort (3 pre- and 3 post-weight loss, non-paired).
